Varietal Identification and Grapevine Germplasm Valorization

The platform guarantees the distribution of genetic material for non-commercial purposes and the breeding of plant material, the characterization of disease resistance at both the phenotypic and genetic level, and finally the varietal identification.

Germplasm bank

The ex situ germplasm collection includes more than 2,000 genotypes (unique genetic profiles), represented by 1-20 accessions per genotype. Accessions differ according to their origin and/or by distinctive phenotypic characters (eg color of the berry).

Genetic and phenotypic database

The FEMVitisDB is being developed to make the list of genotypes present in the FEM grapevine germplasm collection available for consultation. The database collects the genetic profiles and phenotypic characteristics of each accession.

Equipment for plant breeding and resistance phenotyping

We have two large greenhouses and tunnels for plant breeding, as well as growth chambers and mini-greenhouses at controlled temperatures and humidity to carry out artificial infections with downy and powdery mildew.

Molecular laboratory equipment

The platform is equipped with thermal cyclers (GeneAmp 192-well PCR System 9700 and Veriti 96-well Thermal Cycler, Applied Biosystems) and centrifuges (Centrifuge 5804 R, Eppendorf).

Genotyping and sequencing platform

The genotyping and sequencing platform carries out the fragment analysis phase by capillary electrophoresis, returning the raw data which are then processed for the purpose of genetic identification or for disease resistance detection.

Services

Distribution of genetic material

Buds are sold for non-commercial purposes with its MTA. It is possible to book the buds of the accessions of interest within the month of October; the material will be provided by January in the form of shoots of 4-5 buds (for a total of 20).

Breeding of plant material

In the greenhouse we breed seedlings and adult plants in pots of different sizes as needed.

Phenotyping for resistance to downy and powdery mildew

By means of artificial infections on potted plants and on leaf disks we are able to characterize the levels of resistance/susceptibility to downy and powdery mildew at a phenotypic level (symptomatology). Studies regarding black rot are ongoing.

Identification of genetic resistances

By genotyping at 13 resistance loci (containing genes), we are able to characterize varieties or accessions for resistance to downy mildew, powdery mildew, phylloxera and Pierce's disease. Studies on resistance to black rot are ongoing.

Varietal identification

Through genotyping with 9 universal microsatellites, we are able to identify varieties within the vinifera (sativa and sylvestris) and non-vinifera (hybrids) species. Verification of True-To-Type can also be achieved by pedigree analysis.

Applications

Viticulture

The services provided ensure the maintenance and characterization of the genetic variability present in the Vitis genus, with a view to obtaining new varieties suitable for the viticulture of the future.

Nursery

The services offered provide analytical-scientific support in the context of disputes relating to the verification of the correct genetic correspondence of a variety.

Support for experimentation/research programs

The services released support experimentation/research programs that require specific genetic materials (nucleic acid extracts, bud release, plant breeding) or phenotyping tests for disease resistance.
